点击化学利用四嗪(Tetrazine)碳纳米管实现预靶向肿瘤成像

利用TCO和Tz之间快速的正交切割反应,把Tz搭载于肿瘤靶向性的纳米材料单壁碳纳米管SWCNTs上(Tz@SWCNTs),实现对肿瘤的预靶向激活。其主要原理:利用通透性增强与滞留效应,使Tz@SWCNTs在肿瘤组织中积累。通过TCO前药分子或探针,由在肿瘤处富集的Tz进行正交切割。
通过研究tAMC与Tz@SWCNTs之间的释放动力学,相比于mTZ小分子,Tz@SWCNTs对tAMC的衰减反应性更强,且能在生理环境下,对MCF-7癌细胞进行靶向并激活tAMC荧光。
将阳离子染料半菁HCA和TCO相连得到近红外荧光探针tHCA用于通过生物正交释放反应的激活近红外荧光探针。该探针在生理环境下因Tz@SWCNTs的激发快速释放,并在720nm下发出强荧光。
