MB™ 543 DBCO is an orange colored fluorescent probe with absorption and emission maxima at 544 and 560 nm, respectively. The dye is based on a novel class of rhodamine dyes. Unlike traditional rhodamine dyes, MB™ 543 is not only bright and photostable but also highly water-soluble and pH-insensitive. MB™ 543 fluorescent dyes can be used in place of TAMRA, Alexa Fluor 546®, DyLight 546®, CF™543 and spectrally similar dyes.
MB™ 543 DBCO dye reacts with azides via a copper-free “click chemistry” reaction to form a stable triazole and does not require Cu-catalyst or elevated temperatures. In application where the presence of copper is a concern MB™ 543 DBCO is an ideal alternative to copper requiring fluorescent alkynes.
MB™ 543 DBCO reagent is not suitable for staining intracellular components of fixed and permeabilized cells due to high backgrounds.
