修饰聚合物的种类有聚乙二醇(PEG)、聚丙烯酰胺(PPA)、聚乙烯吡咯烷酮(PVP)等,其中PEG是常用的一种。PEG分子与磷脂分子通过共价键结合形成PEG衍生化磷脂,能保护脂质体,使脂质体物理、化学及生物稳定性提高。

Under argon protection, add the compound and tetrahydrofuran to a 250 mL round bottom flask and cool to around 0 ℃ under ice bath conditions. Slowly add sodium hydride (60 wt%, dispersed in mineral oil) into the reaction system in three batches for 30 minutes. Heat the reaction mixture to 40 ℃, and then slowly add 20 mL of dry tetrahydrofuran solution containing 8.35 g (24 mmol) of compound 4 to the reaction system using a syringe. The dripping process lasts for 2 hours. Further stir the reaction mixture in an argon atmosphere for 24 hours. After the reaction is completed, add 200 mL of saline water to quench the excess sodium hydride, and evaporate the organic solvent under reduced pressure. Using dichloromethane (200 mL × 3) Extract the slurry crude product three times, collect and combine the organic extract, wash with 250mL of 50 wt% sodium bicarbonate aqueous solution and 200 mL of water, dry the organic phase with anhydrous sodium sulfate, collect and concentrate to obtain the crude product, and then recrystallize to obtain 6.9 g (6 09 mmol) gray white powder with a yield of 70.24%.
